Professor Gavin Richardson is Lead of the Vascular Medicine and Biology Theme in the Biosciences Institute at Newcastle University. He obtained his PhD from Newcastle University in 2004, studying prostate cancer stem cell biology, before undertaking postdoctoral research at Durham University focused on dermal stem cells, tissue regeneration and skin ageing. In 2010, he returned to Newcastle University on an independent fellowship to apply his expertise in stem cell biology, regeneration and ageing to the study of cardiovascular disease.
Professor Richardson’s research focuses on understanding how cellular senescence contributes to myocardial ageing, inflammation, fibrosis and maladaptive cardiac remodelling. His work combines human cardiac tissue analysis, single nuclei RNA sequencing, spatial transcriptomics, in vitro cardiomyocyte models and preclinical intervention studies to define the cellular and molecular signatures of cardiac senescence. His current research seeks to identify novel biomarkers of cardiac ageing and cardiovascular disease, define the mechanisms linking senescence, mitochondrial dysfunction and senescence associated inflammation, and develop new therapeutic approaches to improve myocardial health in ageing and disease.

Senescence, Regeneration and Cardiovascular Ageing
Ageing is the major risk factor for cardiovascular disease and is associated with poorer outcomes following cardiac injury, including myocardial infarction. Our research investigates why the ageing heart becomes more vulnerable to disease, with a particular focus on the loss of regenerative capacity, the accumulation of senescent cells and the inflammatory tissue environment that develops with age.
Our previous work has shown that cardiomyocyte turnover declines with age, suggesting that impaired regenerative potential may contribute to age-related cardiac dysfunction and reduced recovery after myocardial injury. Alongside this, cellular senescence increases with age. Senescent cells are characterised by a stable loss of proliferative capacity and the production of a senescence-associated secretory phenotype, or SASP, which can promote inflammation, fibrosis, hypertrophy and tissue remodelling.
Senescence and Myocardial Remodelling
We have demonstrated that cellular senescence is induced following myocardial infarction. Through the production of the SASP, senescent cells can create a local tissue environment that promotes further senescence, inflammation, fibrosis and adverse myocardial remodelling. These processes contribute to impaired cardiac function and poorer recovery after injury.
Importantly, our work and that of others has shown that targeting senescent cells can improve cardiac outcomes in preclinical models. Senolytic therapies, which selectively eliminate senescent cells, reduce senescence-associated inflammation, attenuate adverse remodelling and improve cardiac function following myocardial injury. This supports the concept that senescence is not simply a marker of cardiac ageing and disease, but a therapeutically targetable driver of myocardial dysfunction.
Current Research Focus
Our current research aims to define the cellular and molecular signatures of cardiac senescence in ageing and disease. We use human myocardial tissue, single nuclei RNA sequencing, spatial transcriptomics, in vitro cardiomyocyte models and preclinical intervention studies to understand which cardiac cell populations become senescent, how they interact with their surrounding tissue environment and how they contribute to inflammation, fibrosis and myocardial dysfunction.
A key focus is understanding the distinct roles of p16 and p21 expressing senescent cells, how these populations differ between ageing and cardiovascular disease, and whether they can be selectively targeted to improve myocardial health.
Our Goal
Our long-term goal is to develop new approaches to identify, understand and therapeutically target senescence in the ageing and diseased heart. By defining how senescent cells contribute to cardiac inflammation, fibrosis and impaired repair, we aim to develop novel biomarkers and interventions that improve cardiovascular health during ageing and enhance recovery following myocardial infarction.
